Title: Comparison of ?-Amylase producing bacteria between cultivated and uncultivated soil in Faisalabad, Pakistan.
Abstract:
Amylases are extracellular enzymes that degrade polysaccharides such as starch into simple sugars. These are mostly used in pharmaceutical, food, textile, paper, chemical industries, medical fields and clinical research for diagnosis. The purpose of current study was to isolate and compare the α-amylase producing bacteria and their enzymatic potential in both cultivated and uncultivated soil. A total of 40 different soil samples 20 of each both cultivated and uncultivated soil were collected from different areas of Faisalabad in sterile plastic bags. The soil samples were processed by 10-fold serial dilution and spread plate method on Nutrient agar. Bacterial identification was done by colony morphology, microscopic characteristics, including Gram Staining and Biochemical tests according to Burgey’s Manual of Determinative Bacteriology. The α-amylase producing bacteria were identified on 1% soluble starch agar plates by flooding Iodine solution.
